摘要
ObjectivesToestablishamethodforhighyieldmesenchymalstemcellscollection,aswellasaculturemethodforidentifyingmesenchymalstemcellsfromtheswineadipose-derivedmesenchymalstemcell(ADMSC).MethodsSwineADMSCswereisolatedfromfattissuewithcollagenase,followedbyinductionofdifferentiationtoosteogenic,adipogenicandchondrogrniccells.ThesurvivalcurveoftheADMSCatthe37oCand38oCweremeasuredusingWST-1CellProliferationAssayReagent.ResultADMSCsisolatedwithcollagenasefromswineneckfattissuegeneratedastableuniformappearanceafterthesecondgeneration.Thepassageperiodwasfivedays.ADMSCcoulddifferentiateintoosteogenic,adipogenicorchondrogrniccellsunderdifferentcultureconditions.Thehighestgrowthratewasachievedat38oCinthisstudy.ConclusionSwineADMSCshavethepotentialtodifferentiateintoosteogenic,adipogenicorchondrogrniccells,andtheymaybeappropriatefortransplantationforbothresearchandclinicalpurpose.
