Nanotechnology-based combination therapy for overcoming multidrug-resistant cancer

摘要 Multidrugresistance(MDR)isamajorobstacletosuccessfulcancertreatmentandiscrucialtocancermetastasisandrelapse.CombinationtherapyisaneffectivestrategyforovercomingMDR.However,thedifferentpharmacokinetic(PK)profilesofcombineddrugsoftenunderminethecombinationeffectinvivo,especiallywhengreatlydifferentphysicochemicalproperties(e.g.,thoseofmacromoleculesandsmalldrugs)combine.Toaddressthisissue,nanotechnology-basedcodeliverytechniqueshavebeenactivelyexplored.Theypossessgreatadvantagesfortumortargeting,controlleddrugrelease,andidenticaldrugPKprofiles.Thus,apowerfultoolforcombinationtherapyisprovided,andthetranslationfrominvitrotoinvivoisfacilitated.Inthisreview,wepresentasummaryofvariouscombinationstrategiesforovercomingMDRandthenanotechnology-basedcombinationtherapy.
