Theintrinsicgrowthabilityofalltheneuronsdeclinesduringdevelopmentalthoughsomemaygrowbetterthanothers.Numerousintracellularsignalingproteinsandtranscriptionfactorshavebeenshowntoregulatetheintrinsicgrowthcapacityinmatureneurons.Amongthem,PI3kinase/Aktpathwayisimportantforcontrollingaxonelongation.Asanegativeregulatorofthispathway,thetumorsuppressorphosphataseandtensinhomolog(PTEN)appearscriticaltocontroltheregenerativeabilityofyoungandadultneurons.ThisreviewwillfocusonrecentresearchprogressinaxonregenerationandneuralrepairbyPTENinhibitionandtherapeuticpotentialofblockingthisphosphataseforneurologicaldisorders.InhibitionofPTENbydeletioninconditionalknockoutmice,knockdownbyshort-hairpinRNA,orblockadebypharmacologicalapproaches,includingadministrationofselectivePTENantagonistpeptides,stimulatesvariousdegreesofaxonregrowthinjuvenileoradultrodentswithcentralnervoussysteminjuries.Importantly,post-injuryPTENsuppressioncouldenhanceaxonalgrowthandfunctionalrecoveryinadultcentralnervoussystemafterinjury.
