  • 简介:ToaugmentspecificcytotoxicTlymphocyte(CTL)lysisisapromisingstrategyforcancertherapy.Inthisstudy,weexaminedtheboostingeffectofCTLsuponautologouslymphoblastoidBcelllines(LCLs)transfectedwithdiverseplasmids,toexplorethepossibleCTL-basedimmunotherapyofnasopharyngealcarcinoma(NPC).FCManalysisdisplayedratherhighratio(>30%)ofsuccessfullytransfectedLCLsbyutilizingtheDMRIE-Ckit.CTLassaysdemonstratedthatsubstantiallyhigherratioofCTLspecificlysiswasobservedupontheLCLstransfectedwithbothexpressionvectorsencodingEBV-specificepitopesandtheirpresentationmoleculeHLA-A2,incontrastwiththosetransfectedseparately.BytransfectingthevectorencodingHLA-A2alone,onlytheLCLsofHLA-A2+donorselicitedmarkedlyhigherCTLlysis.CTLassaysalsoshowedthatthereexistednomarkeddifferencesupontransfectionbyeitherdifferentvectors(pcDNA3,pNGVL3orpNGVL3-hFlex),ordifferentEBV-derivedpeptides(LMP2Pep1orLMP2Pep2),orwithorwithoutthedoubledDNAsequenceencodingpeptides.ThisstudyindicatedapromisingimmunotherapystrategyonNPCthroughboostingandelicitingtheEBV-specificCTLactivationbytransferringvectorsencodingbothEBV-specificepitopesandtheirpresentationmoleculeHLA-A2intoautologousLCL,thepresentationcellsofMHC/peptidetetramericcomplex.
