Quality Assessment of Transcriptome Data Using Intrinsic Statistical Properties

摘要 Inviewofpotentialapplicationtobiomedicaldiagnosis,tighttranscriptomedataqualitycontroliscompulsory.Usually,qualitycontrolisachievedusinglabelingandhybridizationcontrolsaddedatdifferentstagesthroughouttheprocessingofthebiologicRNAsamples.Thesecontrolmeasures,however,onlyreflecttheperformanceoftheindividualtechnicalmanipulationsduringtheentireprocessandhavenobearingastothecontinuedintegrityoftheRNAsampleitself.Herewedemonstratethatintrinsicstatisticalpropertiesoftheresultingtranscriptomedatasignalandsignal-variancedistributionsandtheirinvariancecanbeidentifiedindependentlyoftheanimalspeciesstudiedandthelabelingprotocolused.Fromtheseinvariantpropertieswehavedevelopedadatamodel,theparametersofwhichcanbeestimatedfromindividualexperimentsandusedtocomputerelativequalitymeasuresbasedonsimilaritywithlargereferencedatasets.Thesequalitymeasuresaddsupplementary,non-redundantinformationtostandardqualitycontrolestimatesbasedonspike-inandhybridizationcontrols,andareexploitableindataanalysis.AsoftwareapplicationforanalyzingdatasetsaswellasareferencedatasetforAB1700arraysareprovided.TheyshouldallowAB1700userstoeasilyintegratethismethodintotheiranalysispipeline,andmightinstigatesimilardevelopmentsforothertranscriptomeplatforms.
